    A Novel Keyword Suggestion Method to Achieve Competitive Advertising on Search Engines

    Search engine advertising is a popular business model for online advertising and recently a new strategy (i.e. competitive advertising) is emerging. Competitive advertising is helpful for organizations to expand market shares from competitors, which is crucial to sustain competitive advantage. To achieve the goal of competitive advertising, appropriate and fruitful competitive keywords should be provided to advertisers. However, existing keywords suggestion methods usually recommend general business keywords based on co-occurrence analysis. They not only fail to enable competitive advertising, but also limit advertisers to a small number of hot keywords, causing high bidding costs. As a response, this study proposes a competitive keywords suggestion method based on query logs. It uses the indirect associations between keywords and the hidden topic information captured by query logs to recommend competitive keywords. Through the method, massive competitive keywords are mined out to help organizations achieve competitive advertising and simultaneously broaden the choices of keywords for search engine advertising. Experiments are conducted to demonstrate that the proposed method could have a good performance than other methods, proving that it can help organizations well achieve the goal of competitive advertising

    Unveiling the Secrets of Collaboration on Video-Sharing Platforms

    In online video platforms, collaborative content creation gains popularity as vloggers exchange social capital to boost content’s appeal and personal visibility. However, existing research lacks rigorous verification of collaboration efficacy in this context. This study investigates the benefits of collaboration on vlogger influence within the online video domain. Utilizing a staggered difference-in-differences approach, we found that collaboration indeed generates positive impact on vloggers’ influence, as evidenced by the significant increases of their fan and like numbers among the audience. However, managing collaboration activities with suitable intensity is vital, as the marginal utility of collaborations diminishes with increased collaboration numbers. Furthermore, vloggers should avoid excessive non-core activities (e.g., uploading photo albums, audios, and articles) post-collaboration, as these peripheral activities negatively affect collaboration benefits in online video contexts. We believe that the findings of our study will provide helpful guidelines for designing more effective collaboration strategies on video sharing platforms

    Incentive Provision and Pro-Social Behaviors

    Individuals’ pro-social behaviors are driven by altruistic and selfish motivations. In this paper we explore how the introduction of external incentives would influence one’s pro-social behavior both in the short term and in the long run. Using a large data set on Amazon product reviews, we design a quasi-experimental approach where we combine a propensity score matching (PSM) and a difference-in-differences (DiD) method to empirically study the effect of incentive provision on reviewer’s behavior. We apply techniques from linguistics, language processing, and machine learning to propose several novel measures to capture reviews’ writing style and quality. We find evidences consistent with crowding-out and overjustification effects. Our study contributes to the understanding of pro-social behavior and sheds light on how incentives would shift individual behavior

    Novel and recurrent FBN1 mutations causing Marfan syndrome in two Chinese families

    BackgroundTo explore the genetic defects of two families with autosomal dominant Marfan syndrome (MFS).MethodsTwo families with MFS were enrolled in this study. The detailed ocular presentations of the patients were recorded. Whole exome sequencing was performed to explore the pathogenic variants and Sanger sequencing was performed to confirm the gene mutations. Segregation analysis among the family members was made and bioinformatics analysis was performed to predict the functional impact of the mutations.ResultsThe main ocular presentations of the probands were increased axial length and ectopia lentis. Using whole exome sequencing and Sanger sequencing, a novel heterozygous missense mutation (c.5060G > C, p.Cys1687Ser) and a recurrent missense mutation (c.2168A > T, p.Asp723Val) were identified within FBN1, which were co-segregated with the MFS phenotype in the families. Evolutionary conservation analysis showed that codons 723 and 1,687 were highly conserved among several species. Functional impact predictions made using several online programs suggested that the mutations were pathogenic.ConclusionWe identified a novel and a recurrent missense mutation in FBN1 in two Chinese families with MFS using whole exome sequencing, and our bioinformatics analysis indicated that the mutations were disease-causing. Our results expand the mutation spectrum of FBN1 and could help us better understand the genetic defects of the patients with MFS

    Progression of radiographic fibrosis in rheumatoid arthritis-associated interstitial lung disease

    Background and objectivesPreclinical interstitial lung disease (pILD) may represent the early stages of rheumatoid arthritis-associated interstitial lung disease (RA-ILD). However, the characteristics, clinical outcomes, and risk factors associated with fibrosis progression in RA-ILD, including pILD and ILD, remain poorly understood.MethodsBaseline data were compared between patients with RA-ILD and those with RA alone. Multivariate logistic regression and Cox regression analyses were performed to identify risk factors associated with the prevalence and imaging progression of RA-ILD, respectively.ResultsAmong the 371 enrolled RA patients, 32.3% had RA-ILD. Multiple logistic regression analyses identified age over 60.0 years (OR 2.22), smoking (OR 2.09), diabetes mellitus (DM) (OR 3.09), mixed connective tissue disease (MCTD) (OR 2.98), serum lactate dehydrogenase (LDH) levels exceeding 250.0 U/L (OR 6.73), and positive anti-cyclic citrullinated peptide (anti-CCP) antibody (OR 2.06) as independent risk factors for RA-ILD (p< 0.05 or 0.01). Among the 98 RA-ILD patients who underwent follow-up for a median duration of 19.1 months, 51.0% demonstrated fibrotic progression on high-resolution computed tomography (HRCT). Multiple Cox regression analysis identified DM (HR 2.03), Disease Activity Score in 28 joints-Erythrocyte Sedimentation Rate (DAS28-ESR) greater than 5.1 (HR 2.21), and baseline HRCT scores exceeding 5.0 (HR 2.30) as independent risk factors for fibrosis progression in RA-ILD (p< 0.05 or 0.01).ConclusionNearly one-third of RA patients in this cohort had prevalent pILD or ILD, and half of them demonstrated imaging progression during follow-up. DM, higher DAS28-ESR, and advanced HRCT scores were identified as independent risk factors for progressive fibrosis in RA-ILD

    Formation conditions and enrichment mechanisms of the Jurassic lacustrine organic-rich shale in the East Fukang Sag, Junggar Basin, NW China: A reassessment based on organic geochemistry

    Chemical composition of sediments is often used to evaluate paleoclimate condition, provenance, tectonic setting, depositional condition, and paleoproductivity. However, the validity of these proxies has long been questioned. The comprehensive use of organic and inorganic multi-indicators in combination when interpreting issues related to terrestrial shales should be advocated. The paleodepositional environment, origin of organic matter (OM) and factor controlling OM accumulation in the Early Jurassic Badaowan (J1b) and Sangonghe (J1s) as well as Middle Jurassic Xishanyao (J2x) lacustrine shales in the East Fukang Sag are reassessed by using organic geochemical characteristics of the OM. Some previous knowledge is updated, and some knowledge is further supported by more evidence. The typical clay-rich shale developed under a lacustrine sedimental environment, and the thermal maturity of these organic-rich shales has entered the oil window and formed economic hydrocarbon potential for the tight-oil and shale-oil reservoirs. The paleoclimate conditions of the study area were warm and humid from the Early to Middle Jurassic periods but were colder and drier after the Middle Jurassic period. The salinity of the water column ranged from freshwater to brackish conditions. The J2x Formation was deposited under oxic conditions, while J1b and J1s formations developed under suboxic and reducing environmental conditions. The J2x Formation OM mainly derived from higher plants was deposited in a terrestrial environment,while the OM of J1b and J1s formations was a mixed OM derived from higher plants and bacteria with little algae deposited under bay/estuary environments alternated with terrestrial environments. It is effective to reflect the paleoclimate by element index and judge the salinity by the updated element thresholds, but it is not effective to evaluate the paleoredox conditions by common elemental ratios and to evaluate the paleoproductivity by Ba in the study area

    Identification of AaAtg8 as a marker of autophagy and a functional autophagy-related protein in Aedes albopictus

    Aedes albopictus is a primary vector of hundreds of pathogens. Strong environmental adaptability and extensive global distribution of Aedes albopictus make it a severe threat to human health. Autophagy is a cellular process involved in maintenance of cellular homeostasis and recirculation of cytoplasm to generate macromolecule constituents and energy under different stress conditions. Many autophagy-related (Atg) proteins have been identified in yeast and were found in various organisms subsequently, indicating that the basic mechanism of autophagy is well conserved in eukaryotes. Among all Atg proteins, Atg8 plays important roles in autophagy and is widely used as a marker to monitor autophagic activity in yeast, Drosophila, nematodes, zebrafish and mammals. By now, Atg proteins in Aedes albopictus have not been reported yet and the autophagy pathway in Aedes albopictus remains unclear. This study identified a homolog of Atg8 from Aedes albopictus and named it AaAtg8. Sequence analysis revealed that AaAtg8 was highly conserved in the Atg8 family. This work proved that AaAtg8 was a functional Atg protein of Aedes albopictus and expressed during developmental and adult stages of Aedes albopictus. Moreover, the study also established the basic methods for autophagy study in C6/36 cells. First, it was proved that both rapamycin and starvation were applicable ways to induce autophagy in C6/36 cells, and that 3-methyladenine and chloroquine could be used to inhibit early and late stages of autophagy in C6/36 cells, respectively. Second, the results in this study showed that monodansylcadaverine staining could be used to detect autophagy in C6/36 cells. Additionally, the study revealed that the level of autophagy in C6/36 cells could be monitored by the turnover assay of AaAtg8 or fluorescent AaAtg8. Taken together, this study identified AaAtg8, the first reported Atg protein in Aedes albopictus. It also provided useful methods for studying autophagy in Aedes albopictus. To our knowledge, this is the first work about autophagy in Aedes albopictus

    Identification and experimental validation of key m6A modification regulators as potential biomarkers of osteoporosis

    Osteoporosis (OP) is a severe systemic bone metabolic disease that occurs worldwide. During the coronavirus pandemic, prioritization of urgent services and delay of elective care attenuated routine screening and monitoring of OP patients. There is an urgent need for novel and effective screening diagnostic biomarkers that require minimal technical and time investments. Several studies have indicated that N6-methyladenosine (m6A) regulators play essential roles in metabolic diseases, including OP. The aim of this study was to identify key m6A regulators as biomarkers of OP through gene expression data analysis and experimental verification. GSE56815 dataset was served as the training dataset for 40 women with high bone mineral density (BMD) and 40 women with low BMD. The expression levels of 14 major m6A regulators were analyzed to screen for differentially expressed m6A regulators in the two groups. The impact of m6A modification on bone metabolism microenvironment characteristics was explored, including osteoblast-related and osteoclast-related gene sets. Most m6A regulators and bone metabolism-related gene sets were dysregulated in the low-BMD samples, and their relationship was also tightly linked. In addition, consensus cluster analysis was performed, and two distinct m6A modification patterns were identified in the low-BMD samples. Subsequently, by univariate and multivariate logistic regression analyses, we identified four key m6A regulators, namely, METTL16, CBLL1, FTO, and YTHDF2. We built a diagnostic model based on the four m6A regulators. CBLL1 and YTHDF2 were protective factors, whereas METTL16 and FTO were risk factors, and the ROC curve and test dataset validated that this model had moderate accuracy in distinguishing high- and low-BMD samples. Furthermore, a regulatory network was constructed of the four hub m6A regulators and 26 m6A target bone metabolism-related genes, which enhanced our understanding of the regulatory mechanisms of m6A modification in OP. Finally, the expression of the four key m6A regulators was validated in vivo and in vitro, which is consistent with the bioinformatic analysis results. Our findings identified four key m6A regulators that are essential for bone metabolism and have specific diagnostic value in OP. These modules could be used as biomarkers of OP in the future

    Highly efficient enzymatic preparation of isomalto-oligosaccharides from starch using an enzyme cocktail

    Background: Current commercial production of isomalto-oligosaccharides (IMOs) commonly involves a lengthy multistage process with low yields. Results: To improve the process efficiency for production of IMOs,we developed a simple and efficient method by using enzyme cocktails composed of the recombinant Bacillus naganoensis pullulanase produced by Bacillus licheniformis , \u3b1-amylase from Bacillus amyloliquefaciens , barley bran \u3b2-amylase, and \u3b1-transglucosidase from Aspergillus niger to perform simultaneous saccharification and transglycosylation to process the liquefied starch. After 13 h of reacting time, 49.09% IMOs (calculated from the total amount of isomaltose, isomaltotriose, and panose) were produced. Conclusions: Our method of using an enzyme cocktail for the efficient production of IMOs offers an attractive alternative to the process presently in use
